  • Organ development is guided by a space-time landscape that constraints cell behavior. This landscape is challenging to characterize for the hair follicle – the most abundant mini organ – due to its complex microscopic structure and asynchronous development. We developed 3DEEP, a tissue clearing and spatial transcriptomic strategy for characterizing tissue blocks up to 400 µm in thickness. We captured 371 hair follicles at different stages of organogenesis in 1 mm3 of skin of a 12-hour-old mouse with 6 million transcripts from 81 genes. From this single time point, we deconvoluted follicles by age based on whole-organ molecular pseudotimes to animate a stop-motion 3D atlas of follicle development along its trajectory. We defined molecular stages for hair follicle organogenesis and characterized the order of emergence for its structures, differential signaling dynamics at its top and bottom, morphogen shifts preceding and accompanying structural changes, and series of structural changes leading to the formation of its canal and opening. We further found that hair follicle stem cells and their niche are established and stratified early in organogenesis, before the formation of the hair bulb. Overall, this work demonstrates the power of increased depth of spatial transcriptomics to provide a four-dimensional analysis of organogenesis. ### Competing Interest Statement A.S. and R.K. are co-inventors of a patent application based on the methods described in this study.

  • Accurate positioning in urban canyons remains a challenging problem. To facilitate the research and development of reliable and precise positioning methods using multiple sensors in urban canyons, we built a multisensory dataset, UrbanNav , collected in diverse, challenging urban scenarios in Hong Kong. The dataset provides multi-sensor data, including data from multi-frequency global navigation satellite system (GNSS) receivers, an inertial measurement unit (IMU), multiple light detection and ranging (lidar) units, and cameras. Meanwhile, the ground truth of the positioning (with centimeter-level accuracy) is postprocessed by commercial software from NovAtel using an integrated GNSS real-time kinematic and fiber optics gyroscope inertial system. In this paper, the sensor systems, spatial and temporal calibration, data formats, and scenario descriptions are presented in detail. Meanwhile, the benchmark performance of several existing positioning methods is provided as a baseline. Based on the evaluations, we conclude that GNSS can provide satisfactory results in a middle-class urban canyon if an appropriate receiver and algorithms are applied. Both visual and lidar odometry are satisfactory in deep urban canyons, whereas tunnels are still a major challenge. Multisensory integration with the aid of an IMU is a promising solution for achieving seamless positioning in cities. The dataset in its entirety can be found on GitHub at .

For this study, we first generated near-annual resolved stable carbon and oxygen isotope (δ13C and δ18O) and element/Ca ratios (Sr/Ca, Ba/Ca, Pb/Ca, U/Ca) time series for 1880–2015 from sclerosponge samples (Acanthochaetetes wellsi) collected at Miyako Island and Okinawa Island in the Ryukyu Islands of southwestern Japan. The δ13C records exhibited a typical variation of anthropogenically derived Suess effects, demonstrating that the rates of decrease of –0.0043‰/year before 1960 and – 0.024‰/year after 1960 in the northwestern subtropical Pacific were respectively similar to and about 1.4 times higher than those of the Caribbean Sea in the tropical Atlantic. Spectral analysis of the δ18O time series revealed significant periodicity of approximately 2, 3, 6.5, 7–10, and 20–30 year/cycle, indicating that sea surface conditions in the southern Ryukyu Islands had been dominated by interannual and decadal variations in temperature and seawater δ18O since the late nineteenth century. The Sr/Ca and U/Ca ratios for the species A. wellsi (high-Mg calcite) might not be a robust proxy for seawater temperatures, unlike Astrosclera willeyana and Ceratoporella nicholsoni sclerosponges (aragonite).
